To better define the processes and population dynamics associated with deep water mixing events in ocean surface waters, we performed controlled experiments using surface seawater microcosms amended with deep seawater samples. Whole, unfiltered seawater from 75m depth was distributed into replicate microcosms (18L each), that were amended with 2L seawater samples from 700m depth. Microcosms (a total of 20L each) were then incubated at in situ surface temperatures and 75m depth light intensities. By amending 10% deep seawater, we estimated a 700-fold increase in NO3, 4-fold increase in PO4, and 5-fold increase in SiO2 in the amended microcosms over the ambient concentrations. Control (20L microcosms with no deep water addition) and experimental (deep seawater amended) microcosms were sampled at 0 hr, 2 hr, 6 hr, 12 hr, 19 hr, and 27 hr time points and extracted for DNA and RNA, and the resulting DNA and cDNA produced was pyrosequenced.
